Actually, nowhere in the game is it specifically stated that Mr. L is Luigi. It's just VERY heavily implied.

Well, for the final battle it was dimentio with the floro-sprout, but before that it was Nastasia who did it because of that whole cutseen with Gary the Goomba.

Thus, Nastasia made him Mr.L right before this level.
