And, in the film, he drives a car, to which the engine has a crankshaft (which is often refered to as "crank" for short). If you look at a car and compare it to a human being, the engine would be the heart. And seeing Statham's character's heart has been poison that it doesn't beat correctly, it'd be like the crankshaft not working.
To use the Wikipedia's description, it is the part of an engine which translates reciprocating linear piston motion into rotation. It typically connects to a flywheel, to reduce the pulsation characteristic of the four-stroke cycle, and sometimes a torsional or vibrational damper at the opposite end, to reduce the torsion vibrations often caused along the length of the crankshaft by the cylinders farthest from the output end acting on the torsional elasticity of the metal.

For example, the ending of John Carpenter's "Halloween", according to Carpenter, was meant to represent that Evil was still out there and was not meant to be an opening for a part two. But the eye-blink for "Crank" and "Remember" for Star Trek II were both excellent details. :)
