Hibana and Jimushi were members of the Oboro Clan. I think they got out before most of the members were killed and Hotsuma went on his revenge Rampage ^_^. Jimushi took Hibana in and raised/trained her in the Ninja. You can see that Hibana wears the Oboro clan insignia on her belt. It is never mentioned if she knows Hotsuma personally but since he was the leader of Oboro I am sure they know of each other in some capacity.

pseudo sequel to be exact. None of the characters in NIghtshade have any direct relation to the characters in Shinobi. Hotsuma is unlockable, and he appears in the opening cutscene, but aside from that its really nothing
