As for it's effectiveness in real life, look at Chuck Liddell's fights in the UFC. He admits to targeting pressure points in his fights, and look where it got him.

The most important thing I have learned in my training from my sensei is that pressure points are NEVER to be used unless you need them. You can easily give just enough pressure to cause pain and prove a point, you don't need to knock out your students to show how cool you are.
Find a SINGLE medical doctor that will tell you this is a safe practice and causes no problems. I dare you.

What's happening is that activating the pressure points send a overload of electric current to the brain and parts of the body and causes a short circuit in the human body.
The excess electric current can knockout a person because the brain suddenly receives a large amt of electricity.
Just imagine a 200W bulb being supplied with 1000 volts. It will BLOW.
Some pressure points do have a lot to do with blood flow. Constricting of the Joints blocks the blood and causes more pain and quickly immobilizes your opponent. Preventing blood flow can be twice as painful as a stiff bat to the shins

However this is common sense. It makes perfect sense too if you have any form of common sense and intelligence yhere is nothing Magical about it.
You stop and start blood flow , or you stop blood flow on two pivotal parts in the body and your entire body reacts. Pull on the webbing under your tongue , that's a pressure point too.
