He is laughing and then out of that laughter comes the statement "You have nothing, nothing to threaten me with.". His hand gestures are portraying that he knows Batman has "Nothing" to threaten him with.

Overall I'd developed a theory that the Joker, at least in this movie, isn't actually insane, but perhaps wishes he was. The interrogation scene was what started me down that road.
