i got lost on :40 where u go up( 6th string) up (5th) up (4th) down up down, down on (5th) after that what do u do its like 5th and 4th u go up up? please helpcoz i got the first part.

Do you mean the main riff at 0:40? That is covered in a previous tutorial.  Try part 1.

Can you describe the finger positions. It is difficult to see from the video

Hi. I'm barring the E and B string on the 10th fret, and the 2nd and 3rd fingers are on the G string on the 11th and 12th frets. You lift your 3rd finger from the G string so you can play the 11th fret on the G string. Place your hand in position for the 10th fret and you should be able to find the notes. Let me know if this does not answer your question.
