It helps to go round the whole tyre periodically to check for and remove glass even if you haven't got a flat, as a lot of punctures are caused by glass that works its way in over time. Just deflate the tyres in the bike every month and go around them with a sharp knife and dig all the glass out.
If you water test a tube for a hole for repair, it's handy to know if the thing that caused to hole is still in the tyre. To help, stick a little bit of mask tape as a marker on the tyre near the valve when you take it off. This way you can relate the position of hole on the tube to the tyre and check it for any glass.
