It will be a totally new protagonist, most of the past ones have been referenced in other GTA games from their own main story. Up until GTA IV theres been something to do with previous plot characters but they've never really had a serious role. Plus, Tommy Vercetti would be like an old man, if it were to be Claude then Rockstar would have to make him a mute, as he is in GTA III and in his brief showing in San Andreas and its clear from the trailer he isnt. Its definitely a whole new character.

You need to remember that Vice City was set in the mid 80's and if GTA V was present day, Tommy would be in his 60's right now. Personally I think it's going to be Claude. In GTA III, we never really got much information about him, and it would be cool to learn more about his past.

Plus if it's Tommy, why would he say he moved to SA because of the weather? It's already great in Miami. If Claude is the main character it would make sense because Liberty City is cold and damp, compared to SA.
