Splinter was originally a normal rat. A pet of a martial artists expert.

When Splinters 'owner' was killed by Shredder, he himself fled. eventually meeting the four turtles. Brushing off the slime they were covered in, he soon realized that he was mutating as they were.

And such begins their adventure. At least according to what i know from the cartoon.

I think the new 00's cartoons play with the movie's version as well (Though I didn't like the 00's cartoons so I never really watched them). I'm not 100% certain, but I'm pretty sure the comics also had Splinter as being a man turned rat.
