No. They shot hours of material for each episode, but it's unscripted. Besides having years and years of practice, if you watch enough improv from the same people, you can begin to see things they use to help themselves on the spot, particularly with singing/rhyming. It's still ridiculously impressive to watch.

Haha, I like how Wayne gives Steve a perplexed courtesy smile at 1:30 after his rapping ran aground. Colbert has no real comedic talent - I don't find his show funny at all. He basically recites lines for an awkward and forced half hour. I much prefer someone like Craig Ferguson, a genuinely funny person who doesn't have a script at all for his monologues.
