A close up just means that the camera is very focused in on someone's face, in this case its Jared and Jensen.
Kripke was saying that Jared was going to have a sex scene, and Jared was saying it was the first time he was hearing of it. He said it wasn't like Kripke said "Hey your going to have a kissing scene, get prepared". Kripke just sorta blurted it out in a somewhat crude way. :)
The look on Jensen's face after Kripke tells Jared he's going to have a sex scene KILLS me every time. He's like, "Whooaaaa."
