The blood layer only consists of the part of the animation, where the blood is running down the hole. As the blood layer is below the blue whole layer, it doesn't need a hole. It just needs to be white where you need full transparency and red where the Bond footage needs to be tinted in red.

I suggest you have a look at my Quick Video Tutorial for the Goldeneye gun barrel. It's practically the same here, though a lot easier as there is no horizontal barrel motion to correct. Still two layers here, one for the blood and one blue color key. You only have to resize the actor's layer according the barrel motion and add the strobe effect (it's a simple invert/negative effect).

I use a white backgground for the bloodlayer. Applying it with the blending mode "Multiply" makes it as transparent as it needs to be.

I'm more impressed with your version than the CR one.
The CGI version in CR looks like a battleship gun rather than a pistol.
It has too many rifling grooves.
