There are two ways (Ok more than two ways but I use these the most) to kill in one hit. You could either mod a weapon that caused some great damage. Or you could open the console, click on the character you wanna kill in one punch and type: SetAv Health 0. This code will give him 0 hitpoints but will not kill him.
