I am a medical student and I did study hemoglobinopathies and thalassemias in first semester. It is true that Sicily, Greece, and Cyprus have sickle cell AS WELL AS beta-thalassemia, but that is not proof of black ancestry.
HbS comes from the 11th chromosome. There are 2 genes for beta chains; at least 300 mutations thus far discovered, some spontaneous.

In fact I paraphrased a study on the topic which stated that the HbS of common-day Sicily is identical to that of African-Americans (another mixed group). The report stated that this one genetic marker demonstrates the proof of African ancestry.
