Is it possible to reach the end without getting all five chests, thus losing one character's equipment, all the money, or all the items forever? Or is there something in the game that only allows you to proceed once you get ALL your stuff back?

You can, indeed lose almost all of your stuff here, if you are foolhardy enough to go without it. I have done it, though it was a long time ago. I think there may be one thing they require you to find before you can go on, but I do not remember what it was, and I could be wrong anyway.
