the liquid is a broth used to stabilize the bacteria, its already made by manufacturers. you can take a throat swab and place it in the broth so it can be stabilized then use the broth for culturing
@huyeniee yes if you have colonies on agar you touch the colony and then tranfer it to the other 'clean' agar plate.. you can use this technique if you didn't get a pure culture first time around
