Should of Vanish/Doom is ass. Unless he is one of the very few enemies in the game who is resistant against vanish!

How do you make gigantuar appear? I walked around the desert for quite long and only slagworms and cactuars appear... Gigantuar never appeared, is there anything I have to do to make him appear?

You should beat 10 cactuars and after that he'll appear in the southern part of the desert.
