It's a total fake. I remember that episode:

The road runner actually gets through that painted street as if it was real. The coyote keeps watching it while it disappears in the distance.

The coyote can't believe it, and looks at the "camera". That's when a truck coming in the opposite way along that fake street, runs him over. That's it.

So they must have done some kind of animation trick over that scene. If you look carefully, the way they move is pretty "robotic", not really fluent.
