I use ca65 (part of cc65, which is a C compiler). It's a nice assembler, but I would recommend another assembler called asm6 to anyone who is just starting out.

@Thepiratejoe Retrousb.com has carts you can purchase there, along with some pretty cool new nes goodies. As far as creating nes games....holy schnikees its complicated. Type in (nes game development) in your browser and prepare to be confused. Its another language bro.
