And do you have to do them all at once?

You need 50 fares. You can do as many runs as u want and save the game, the counter does not reset.

I was doing the taxi mission for 2 hours straight.

Literally 2 hours and the game never said "Taxi Mission Complete"

I left the car twice to eat and that was it.

Maybe I should have kept count.
