That S-Video port is intended for output, usually to a TV. Look for a yellow port on your video card, that's the one you connect to, to capture the video.
Video cards that have on board capture units are ATI AIW(All in Wonder) cards
Most computers have high quality mic inputs but you might as well do it right and that would be to use the line in port witch tends to have a higher range.
