the tuning is how the guitar is tuned. example is standard tuning.

the key has to do with what the song is in. this basically has to do with what chords/ notes you use. the tuning could be standard and you could play in the key of C major(say, using the chords C, Dm, Em, F, G, Am, Bdim) or you could play in the key of E while using standard tuning (using the chords E, F#m, G#m, A, B, C#m, D#dim.)
