that's an incredibly difficult thing to do for arbitrary materials, with arbitrary hole sizes under arbitrary lighting.

could be color based or shape based, you just scan the image with a mask and check for average pixel difference between the mask and the webcam image, depth control would be hard to do, with through holes would be doable. YOu can just mark it with dots though, and scan for a threshold color with a margin of error, anything darker would count as a hole, and you can take size into account too.
