I think what he means is when he says "Boo!" at 0:39 is it possible to use a star right when he says it.

During phase 2 if proper punches are still in this version of the game the same way they were in the NES version, then making sure you hit him with the opposite fist of the one he tries to hit you with when you dodge his blow should give you more damage and therefore a faster time in phase 2.

Just enough damage to take him down with, wow, 2:14 left! Do you think sub 50 is possible with this method, or could something change to speed it up?

This is probably the fastest way outside of some sneaky star gathering for phase 2. sub 50 should be possible this way though, I was slow on my BOO star and the ensuing dodge afterward. One unstunned "frame" in this game seems to be .06 so yeah, under 50 definately possible with this strat.
