It's not good for the neck to do each string consecutively from one side since there is a lot of tension coming from tightening the strings. Doing it from both ends help prevent neck tweaking, string breakage and other stuff :)
Yes it does for 3 on a side gears. The string should always come off the inside of the tuning post. Six in a line technically, not as important. Its more a tuning habit. If you look at the string angle from the nut slot to the post and keep that line as straight as possible, you'll find consistency in the direction you turn your tuner.
How much slack should the string have right before you start wrapping the string around the tuning peg (after you've threaded it through the tuning peg)?
Try detuning your strings until they're real flappy so that you can fit your hand through the sound hole. Reach in and push the bridge pin out with your finger.

I like to give enough slack to go over the string once and under it twice. This way there will be enough tension, holding the string in place.
