Shamisen tuning response, pt. 1:

There are a few standard tunings used, but the most common tuning is that of a fifth and a fourth (from lowest to highest string). There is no specific standard key; I tune to whatever the key of the song happens to be. Or, if the key of the song happens to be above or below the tuning range of the instrument (the tuning range is G to D, or thereabouts), I'll just tune to the 5th of the key.

Shamisen tuning response, pt. 2:

Basically, if the song is in C, I'll tune the strings (from lowest to highest) C, G, C. If it's D, I'll tune the strings D, A, D, and so on...
