In my comment I meant that often two characters are portrayed by the same actor, but the point is that here he mention his other role. So Full House is a TV series within Step by Step. Steve Urkel appearing in-character means that he also exists in that universe and when they watch Full House and see Urkel guest starring, is simply a real-life person having a cameo to them.

The only other possibility is that that John Stamos statement was a fourth-wall breaking and should be ignored...
