Her hologram projection and AI intelligence matrix is based on the flash cloned brain of Dr. Katherine Halsey, who was in fact in her late 50s when Cortana's programming was written--although, she was given the appearance of Halsey of when she was in her mid 20s rather than her actual age at the time.

In the dashboard, go to your console settings. Then go to language settings and change to 日本語. Put in Halo 3 and play.

To save you the trouble. When you go to switch back to english, if you are looking for the language option. Look for the word 言語

I don't know about the single player, I never play it. It should do it, it does it in multiplayer. But I speak both languages and the English voice sounds better for xbox games, so I leave English on most of the time.
