Ada was Leon's, for lack of a better word, sidekick in RE2, having met her in the midst of the outbreak. Throughout they help each other (well, he does all the work, really), and towards the end of the game, its revealed that shes a spy shortly before apparently being killed.

That's the professional connection. ACTUAL personal connection is vague and...well, convoluted enough to the point where everyone has a different take on it.
