If you watch the entire episode you can see that Spock ramps up his own sex appeal as soon as the women beam on board. And yes he is amused at the humans reaction to Mudds women, but he is fighting back with all the eye-sex he can muster...adorable 'non-emotional vulcan' ;)
Also, @ charmolution, jackass wasn't really considered a swear back then. You see it in old cartoons sometimes too, signs calling people jackasses and stuff. I think it meant more like "stubborn brat".
In my opinion Spock isn't smiling to himself. He begins to smile in a barely visible (and adorable) way when Kirk arrives, and in an almost affectionate way he continues talking to Kirk about a critical situation for the ship and for him as captain . No, I would not say that Spock smiles to himself, but to Jim while giving him bad news softly as he can! This scene recalls me "the companion" in "metamorphosis": changes color when he speaks with him! ^_^
