If the song makes it so that they are brothers and sisters then they will be, if the song makes them lovers, then they are lovers. I hope this helps clear things up. ^^

From what I can gather, the samurai fell in love with the priestess. Then, for some reason (probably ordered too), he had to kill her. This breaks her heart, but she seems to accept it as her fate, remembering all the good times they had during her last moments of life
