I think he meant the "center" of Harlem; except here, in Manhattan, downtown refers to the southern part of the island, and uptown refers to neighborhoods in the north - while in the rest of country they usually call the middle (and busiest) part of a city "downtown." (Of course, New Yorkers just HAVE TO be different. :))

It's one of the most famous musical venues in the U.S., some of the biggest African-American singers/bands started their careers here from the Jackson 5 and James Brown to Jazmine Sullivan to Stevie Wonder.
