This is a clandestine video taken at NY or boulder. At the Paramount Theater in Seattle it was specifically requested not only that camera equipment not be allowed, but implicit that nobody make personal images or recordings. What is so difficult about respecting the wishes of someone you've theoretically come to honor?
I guess just the translator found it funny, and then everyone else thought it was funny that the translator was laughing. (I didn't find it particularly funny either, but HH Karmapa's reaction, and the audience reaction were rather funny!)
O yah! I remember this day haha.. It was pretty funny. But the funniest was when the Tibetans would be laughing after HHK tells a joke and all the rest (westerners) would be quite but after when the translator translates what HHK had just said, all the westerners would be then finally laughing.. I don't know.. it's not funny when I write it.. but it was funny hahaha

Sometimes things are lost in translation -- a play on words, for example, or a cultural reference that's not really explicable...
Its possible that the joke was simply something that didn't translate.
I think he was also having a laugh at the Western "picture" of a dharma practitioner...
