As itisnick wrote, technologically speaking, Vocaloid 2 is a refined version of Vocaloid 1. But the design concept of this "Character vocal series", of which Miku is the first one, is a bit different from other Vocaloids.

The sampling source of Miku is an voice actress of anime, FUJITA Saki, who is not a singer. According to the producer of this software, they wanted to give a "personality" and "body" to this voice synthe, so that "she" would be like a "japanese idol" or an anime character.

I don't know about Vocaloid 2, but Vocaloid 1 was a real singer who sang all the different vocal sounds and Vocaloid software connects the right sounds at the right pitch together. So it's closer to a "smart" sampler with a great big bank of one person's edited and processed singing, than a computer generating singing through pure synthesis
