I don't see the problem with this mission. I accidentally killed the Doctor on my first attempt, so I came here to see if pickpocketing was the solution, which it was. Then I restarted it and easily succeeded with 15 seconds to spare.
I completed this with 6 seconds to spare, thanks to a combination of horse riding and free running (as well as some blind luck and a lot of button mashing).
I love Assassin's Creed, but really, really HATE the timed missions.

here a TIP use the horse steal one dont bother calling your one you will only call the prostitutes cos they too close when on the horse just look the map and dont! keep moving when you get to the the doctor jump off your horse stop use a smoke bomb steal the antidote then jump back on thehorse and leg it back you should make it horse saves alot of time dont try roof tops unles you knbow exactly how to get ther with out a hitch
i think i know what the problem is. the 1st and 2nd time i did this i did not get full sync but the 3rd time i got full sync. the 1st and 2nd time there were borgia guards near the doctor but the 3rd time there weren't any near the doctor. I dont think the way you travel to the doctor doesn't matter. going on roof, on horse, and on foot i tried all three ways and they worked without borgia near the doctor. I hoped this helped.

@missyepyep - don't know if anyone responded but NO the two quests are not related. If you can't find Claudia (she should be next to the desk to the right of the stairs just as you enter the door.) then you either have a bug or "The Plan" hasn't advanced completely. I'd suggest going back to the hideout to see if there is still some business to conclude. Otherwise I don't know what to tell you except maybe to go back through DNA and improve the synch. Good Luck.

Thank you SO much! I literaly tried this 50 times and I couldn't get it. I tried going on the roofs & I couldn't get there anywhere NEAR the goal time. It is really messed up that running throught the city is faster than the rooftops. It doesn't match with the logic of the game.
You can immediately grab a horse at the start of this, steal it and follow the route to 0:45 - be in high profile mode and stand on your horse, that way you can jump to the ledge, to hops over to the right on to another horse. Again at 1:20 high profile stand and jump on to the house, smoke bomb the doctor and steal, re-bomb the crowd and climb back over the building and jump onto your horse, ride it to the previous horse your left as you stand and leap over, or call a new horse. Easy run now!
I Had a Better Run. I took the Same route except some less jumping, and when you get to the Doctor throw a smoke bomb so the Doctor and the Guards don't catch you then go do the same route back, i hade like 31 Seconds remaining :P my first 2 tries didn't go out so well cuz i forgot i had smoke bombs :P
I f*cking hate those specific objectives that you have to do to get 100% sync on each mission because most of the time it makes Ezio seem like a complete retard instead of the master assassin that he (supposedly) is. Ubisoft fucked it up to the point where it isn't even fun.
@2287nemo the 100% sync objectives make it alot better, they actually make it interesting by adding some difficulty at times,as otherwise the whole game wud just be stupidly easy, not that it's isn't already but more so, besides there optional, so if u reli want u don't have to bother with them u just won't get the repress memory sequeces at the end!!
@DaRkLoRd1061 whether the 100% sync objectives are good or bad is a matter of opinion.. I think it sucks. And yes it is optional but like many other gamers I'm a bit of a perfectionist (have to get all the trophies, have to work out every little hidden thing, all that stuff) and having a game that says "99.8% complete" has failure written all over it.
@DaRkLoRd1061 and whether the objectives make it harder or not is also a matter of opinion. I found some missions very difficult (spent more than 5 hours on the tank mission)... Oh, and I just had to start all over again because, like many others, I was 'foolish' enough to sell the first shrunken head I got because I didn't know you were supposed to use it for the armour later because the game doesn't disclose that information till much later.

I already played the first two AC's and I always thought taking to the roof tops was the fastest route. On this mission taking a horse, and or running on the ground is by far your best option. Stay OFF the roof tops unless absolutely necessary!

@em2skinner8 well you have to understand that not all the rooftops are level. you have to run up walls and climb them which takes a significant amount of time.
@em2skinner8 I know. I tried doing the rooftops, and kept failing. Seems stupid that running through a maze of streets is faster than a more or less direct route across the roofs.
@em2skinner8 normally the rooftops ARE faster, but in this area specifically you have to take into account the fact that you have to scale a building and the fact that the spaces between buildinigns are too far apart an you ave to constantly get a new grip and climb up the walls again.

@em2skinner8 well you have to understand that not all the rooftops are level. you have to run up walls and climb them which takes a significant amount of time.

@em2skinner8 I know. I tried doing the rooftops, and kept failing. Seems stupid that running through a maze of streets is faster than a more or less direct route across the roofs.

@em2skinner8 its faster for roooftops for me,but it's alot harder to run across the rooftops because of the mixed building sizes/completions :\

@em2skinner8 normally the rooftops ARE faster, but in this area specifically you have to take into account the fact that you have to scale a building and the fact that the spaces between buildinigns are too far apart an you ave to constantly get a new grip and climb up the walls again.
