This... bored the crap out of me for the first half. Much like the actual game itself. To that end, this is certainly more representative of what the game is like.
Its a shame 3DR and Triptych got pushed to the back seat in a way. This trailer is fantastic. Contains the air of the old guard in a way. I think the music could be a bit more beefy when the action picks up, but I love it nevertheless.

Look at how Gearbox advertises Duke now. They treat him like a gangsta rather than "The Ultimate Ass Kicker." I personally believe that rap does not go well with Duke Nukem at all. It feels like GBX is trying to appeal to the Jersey Shore audience.
Also, notice how the trailer is devoid of any Gearbox logos? Triptych is a group of former 3D Realms employees too. Last, notice at the end of the video it says "Spring 2010"? 3D Realms intended DNF to go gold on April 1st, 2010.

Look, play Duke Nukem Forever, if you hate Gearbox so much, pirate the damn thing, but I am telling you man, I 100% guarantee that if you play through Gearbox's Duke Nukem Forever for at least 2 hours, you will see gameplay that looks frame for frame exactly like the majority of this trailer.

All Gearbox really did was finally fund the damn thing well enough to get it in a state to be released instead of filled with bugs you'd expect from a beta, 3D Realms did all the hard work though so this is really their game. All those Gearbox critiques are 3D Realms' fault, not Gearbox.
I like the model used for duke here a lot more than the final one.
That said, every other graphical change is pretty good, no complaints there.
This is how a DNF trailer SHOULD be though, gearbox completely screwed that up.
