It's kinda goofy that she's using a microphone during Koi wa Sensou when she already has a headset, but I guess since Koi wa Sensou isn't a song that can accomodate a cutesy choreography, they needed her to act like an actual singer does on an actual stage, so she had to hold a microphone or it would just look awkward.

There is no one singing behind a microphone when Miku's singing. It's actually just a synthesized voice created by altering voice actor samples.
