Yeah -- if you already played the stage and got a gold card or two, you can replay the stage and get any gold cards you missed.

With bosses, you have to get all 5 gold cards in one fight. If you get, for example, 2 cards in one boss fight, you can play the fight again to get all 5 gold cards, but you have to earn them all in one fight, you don't get to keep the ones you got before.
