Bloodlines actually deals with how the characters are related to eachother--two characters have been revealed to be related. Also, it deals with the special qualities of the blood of several characters. It's actually a pretty accurate title. Sorry it wasn't what you were expecting.

In Part 3, we see literal blood lines from Emma to the Elder during the ceremony, as we saw the season 1 finale with Bree.

If you watch Part 4, you will see that there is a twist that involves two characters who turn out to be related. DNA and genetics are definitely a subtext if you follow the story.
