After awhile the slashes were dropped as the meme spread beyond 4chan, and the "male-only" interpretation comes from the fact that (i believe coincidentally) 'brony' is a portmanteau of 'bro' and 'pony'.

The purpose of the word is to get across a certain idea. When you have a slang word, it has its own rules. While "bro" is usually a male term (though I as a female have had it said towards me), if someone calls someone a brony you know they mean that the person is a fan of the show.
