I think from all the lost seasons this part in Season 4 part 5 when explains all the story between the Desmond and Penny it is so strong and its something which will not happen everyday and in every move there is something special in all in text, conversation the scenario and the best of the best acting part is the call conversation when the promise to each other that they will do everything to find each other. I love this part.

According to this episode and based on some information about the show(that it has something to do with stephen hawking and quantum physics) you can see that all realities(past,present,future) are happening at the same time.You can confirm because in the moment that Penny answered the phone, Desmond puts a big smile on his face(both Desmonds-from the "past" and from the "present")

It can be had from the Season3 soundtrack / disc 1 -Track 14 "Distraught Desmond" about 1:20 into the track the soft piano begins and since you're looking for it, the tears too.

Distraught Desmond was used in Desmonds episode last season. The version they used in this scene was like a reprise, therefore parts were different, like the way it picks up when he says "i love you." So yeah, this version of the song ain't out yet unfortunately.

Even though the show has really taken off from where it started, gone away from it's originality and become... Well, WILD, this episode was one of the best episodes of any show ever, and I really was moved by it! Incredible... They did it so well. The call was excellent and had such a good feel... Nothing else to say about it. Desmond/Penny-story is one of the best parts of the show. Maybe even THE best.
