I think that really depends on your existing picking style/technique - could be minutes, could be days. I also find its easier to get one when you are learning if your pick hand is more parallel to the top of the guitar with your thumb inline with your wrist - than a classical style where your pick hand is more perpendicular to the strings.
Its about 11:30 so i dont feel like pulling a amp out to try this out, but do you think ill be able to do this with a 1959 melody maker with only a bridge pickup?
People saying they need this or that are incorrect - pinch harmonics can be done on an acoustic, all over the neck, if you have correct technique. Electronics often inserts artificial harmonics which makes it easier to do when starting - but isn't necessary once you have the technique down. So... no, you don't need humbuckers or high gain amps or pedals when it comes right down to it, those things just make it easier when learning.
guitar brand will make no difference - but certain guitars are more harmonically active, usually from high end neck construction - but that shouldn't make a difference when learning.
would it matter what kind of amp i use?? because the one i have now sucks... when i do this i can hear it but barely.. it's only a 10 watt so yea that might be my problem lol
that wouldn't be your problem... technique is technique, regardless of equipment - but - in this case, the volume of your pinch may come through better using a distortion pedal as you improve upon technique

no - firmly fretting the note will produce a 'pinch' harmonic, vibrato or a bend will add sustain/color to the harmonic though.
