Well... it's not easy to explain, but basically, Jacob (you do know who Jacob is, right?) has a brother simply dubbed the 'Man in Black' and, in a nutshell, Jacob was the protector of the light in the island, the light that keeps everything in the world alive, and his brother became kinda evil, until Jacob killed him and sent him down the tunnel thing to the light source, which for some reason his adopted mom told him never to go down. If the lock had been on the outside. When Charlie locks the door he does it with two handles on the inside. Des can't open it because there is a weel on the outside but it's not a lock. There was no way to lock it from the outside. I'm not sure how Charlie knew this, but they did want Desmond to live.

Well, noone really knows what it is yet. It appears to be some kind of living smoke who wants to kill people, but what it really is and why it kills people is still a mystery. However, there are some theories about it. One of the theories is that the monster is a defense system of the island. A map in an episode a long time ago said something about the "Cerberus defence system", and that's propably what it is. But noone, except the creators, knows more than that. :(
