Not necessarily. The original work called it cosuppression because it was consistent with other reports at the time of gene silencing upon overexpression. RNAi is one way that this is achieved, but there also are hard changes in DNA (like methylation) that occur as well to silence the process at the level of transcription. At the time of Napoli et al it was unknown what the mechanism actually was, but it was clear that something revolutionary was happening!
