Nel linguaggio parlato le due cose si confondono 100 volte su 100, ma dire, per esempio: "lui/lei è bella" è teoricamente un errore... bisognerebbe dire "egli/ella è bello"... nel linguaggio scritto è bene tenere presente la distinzione.

"Egli or Ella " are normally (p.e. Newspaper)used as Subject but "Lui or Lei" are for both subject and object.
"I say to him" is "dico a lui". "Dico a egli /ella" is wrong. Ella and Lei are used as Subject and Object. Ella is older then Lei. Esso or essa is used for things and animals.

This is the rule, but in the spoken laungauge we use lui/lei both as subject and complement. So,when you are speaking,always use lui/lei or you'll end up really weird and funny to an italian ear.But when you write you better use "ella/egli" as subject and "lui/lei" as complement or you'll wrinting will be rough
