Good question. It all depends on what part of the country or you are from.

In California we are more likely to pronounce the O. Also, people who are engaged in the professional study or care of this marsupial will probably pronounce the O. Most people simply say possum. Those who do rehab and get all silly about the sweet creatures are often heard to say possie. It all works. Just be nice to them.

In California you will frequently hear "O possum", especially by people who tend to them. According to most dictionaries both pronunciations are correct. It's interesting how much this affects people. So much so that in my latest videos I divide the pronunciation equally.
