Have you watched the series? If you haven't, it probably won't make sense since you need to know Dexter's character. And if you have watched the show, well... It should make sense then.
It's funny because anyone who watches the show knows that Dexter would never kill the yoga teacher, but she's so annoying that we can sympathize with his momentary temptation to do so. ;-) Plus the look on his face is hilarious!

@K4zKyzah You can't really explain comedy. If you don't get it, you don't get it. Someone can tell you why they think it is funny, but it probably won't make it funny for you.

Its dark humor. Its funny at something it shouldn't be. Hope I explained that right.
And I agree with Emdier. Funniest scene ever. The look on Dexter's face at 00:37 is priceless.
