It's not just people who HAVE done a human transmutation, but people who have the potential to do it, since it's an extremely complicated procedure requiring a lot of intelligence and skill.

Roy has both the drive and willpower to do it, and eventually is forced to do so at some point in the manga.

The only reason Al couldn't do it from the start is because when he first saw Truth he was traumatized and so blocked out the memory, therefore he blocked out the knowledge of how to perform Alchemy without the transmutation circle. Once his memory returned the knowledge returned with it.
