1) GLaDOS was voiced by an actual human singer, then made more "robotic" in post-production and

2) SO WAS THIS SONG

@vidmuncher Er...yes and no. An actual human provided a zillion different voice samples to create the Vocaloid program, which is in turn used to generate Miku's singing voice. GLaDOS, on the other hand, had her lines voiced directly.

Well, to answer your first question, Reimu is from game series called Touhou Project, where she acts as main character. But because of the wide variety of fan comics based on Touhou, it's common for unaware people to mistake it as manga series
