Like some people pointed out, the joke is about ninjas being masters of stealth, hence his looking for ninjas in the room. When he, at the end, runs in front of the mirror looking at it, it's because he's figuring the guy in the mirror (i.e. himself) might be tricking him. Pretty funny.

All aside, why would ninjas mimic someone facing the opposite direction (00:36) and lose track on the mimicked?
